![]() The Base 2 project run by the Northern Ireland Association for the Care and Resettlement of Offenders, which works for the reintegration of individuals into the community, shows that the number of referrals to NIACRO has increased steadily year on year. That is far from the case, and some statistics show that the opposite has happened. They may think that punishment beatings, expulsions and evictions by intimidation have disappeared. Most people think that, with the ceasefire and the ending of violence in Northern Ireland, many problems have gone away. That disappointing delay can be explained partly by the intervention of the election and the summer recess, but eight months cannot be entirely explained away. ![]() Disappointingly, the Government did not respond to our report until the beginning of December, eight months after the report was published, and well beyond the deadlines that Government set themselves. The inquiry was launched in 2000 evidence was taken then, and at the beginning of 2001. During this Session, that was printed with the Government's response in the fourth special report. I am happy that the executive supported the Committee's inquiry, and provided a response, for which we are grateful. One problem with the new devolved institution in Northern Ireland is the involvement of the Northern Ireland Housing Executive in providing support for victims who have been removed from their homes. The report inevitably touched on devolved matters. I am trying hard to follow his difficult act. I pay tribute to him on behalf of my colleagues for the sensitive and light touch with which he handled the Select Committee for five years-I am not allowed to notice him in the Public Gallery today. Friends know, but not everyone will know, that the report was prepared before the last general election, and therefore not under my chairmanship but under that of my noble Friend Lord Brooke. It is important that the subject is aired, and if we cannot air it on the Floor of the House, it is as well to do so here. ![]() First, I thank the Liaison Committee for allowing the debate. ![]()

Cartagena and Forysthe said evoking that feeling of poring over the map and imagining the world of Middle-earth in the reader’s mind was one of the team’s very first ideas when concepting the set. Less flashy but nonetheless iconic are the basic land cards for Tales of Middle-earth, which trade panoramic vistas for sections of a map a reader might find printed in the back of an old copy of the book or Free League’s The One Ring tabletop RPG. Characters printed in this style will reportedly be captured in a moment of internal struggle or hardship. Cartagena said the stylised treatment, which depicts the wizened member of The Fellowship’s fateful battle and fall against the Balrog, is meant to communicate what’s happening “inside the Ring” - hence, the circular frame and ancient Elvish text. One more Gandalf is likely to appear, given Dmitry Burmak’s teased artwork titled Gandalf, Friend of the Shire, but we know for sure that Gandalf the Grey can pop up bearing Tales of Middle-earth’s showcase frame treatment. ![]() Much like book Gandalf, the card has an exit and return baked into its very nature. All four abilities can only be chosen once, which means Gandalf, bearing the sword Glamdring in Aaron Miller’s artwork, must exit the battlefield before pulling again from his bag of tricks. Any time Gandalf’s controller casts an instant or sorcery, they may choose from among four options: tap or untap a one permanent, deal three damage to each opponent, copy a spell they control or put Gandalf on top of its owner’s library. Not so for Gandalf the Grey, whose card bears a dual red and white Mana colouration and abilities befitting the archetypical fantasy wizard. That shift is translated through the artwork for the as-yet-unrevealed card, though the interview heavily intimates its colour identity will match his title. The irascible but doting grey wizard is maybe the best example, as his death and eventual rebirth as Gandalf the White marks a drastic change in both his demeanour and function within Tolkien’s story. ![]() Gandalf’s card will actually be one of at least three versions of the wizard to appear in the set, an intentional choice that allowed the designers to portray central characters at different points in the narrative. Watch on YouTube Wheels teaches you everything needed to start playing Magic: The Gathering. It then draws cards when tapped equal to the number of burden counters but also drains that much life from its controller - this dual-edged sword mirrors the manipulating effects The One Ring displays on Smeagol, Frodo and other bearers. The indestructible legendary artifact provides its caster protection from everything until their next turn, as long as it was cast and not tutored out or otherwise sneaked onto the battlefield. The One Ring, which Frodo carries from The Shire to the fires of Mount Doom, will appear in full glory on its own card, and it’s appropriately powerful. Their insights accompanied two revealed cards demonstrating some key mechanical insight alongside the now expected showcase frame treatment. In a recent interview with IGN, Art Director Ovidio Cartagena and VP of Game Design Aaron Forsythe discussed the team’s approach when synthesising one of the most well-known fantasy stories ever into the five Mana colours and mechanical limitations of Magic: The Gathering. RESULTS A large proportion of CRISPR dark matter spacers can be annotated by pangenome analysis In addition, our results demonstrate that the study of thousands of genomes of the same species allows us to reduce the CRISPR dark matter and to trace the origin of most of the spacers found in them. This has motivated us to investigate whether this relationship could be mediated by phages that could take advantage of the existence of these membrane proteins as receptors or adhesion sites to infect bacteria. Then, these genes have been functionally analyzed, and we found that they are enriched in genes encoding membrane proteins. We have created a large pangenome for each species and compared strains with or without CRISPR-Cas systems to discover genes associated with the first group. ESKAPE bacteria have CRISPR-Cas systems of the most common types, from I to IV, but only in a minimal number of strains, with frequencies ranging from less than 1 to 60% of genomes, depending on the species ( 16). In this work, we analyzed thousands of genomes of the group of bacteria known as ESKAPE, whose acronym refers to two Gram-positive bacteria ( Enterococcus faecium and Staphylococcus aureus) and four Gram-negative bacteria ( Klebsiella pneumoniae, A. There are different types of CRISPR-Cas systems based on genes that are part of the different steps of this immune system (adaptation or spacer integration, expression, and interference) and are generically called cas (CRISPR-associated) genes. They provide acquired immune resistance against phages and other foreign nucleic acid molecules such as plasmids, thus restricting gene transfer ( 4). CRISPR-Cas is an adaptive immunity system found in most archaea and in less than half of the bacteria sequenced ( 3). Restriction-modification systems are by far the most abundant, being present in 83% of prokaryotic genomes, followed by CRISPR-Cas with about 40% ( 2). Bacteria defend themselves against infection by these phages by means of different molecular strategies. They can be used to control bacterial growth and are even beginning to be used to treat infections in humans ( 1). To our knowledge this is the first example of gene therapy for an autosomal dominant gain-of-function disease using a disease allele inactivation strategy in place of the less efficient disease allele repair approach.Bacteriophages, also known as phages, are viruses that predate bacterial cells, representing environmental burdens for their growth and spread. The WHIM allele-inactivated HSCs retained long-term pluripotency and selective hematopoietic reconstitution advantages. Then, through in vivo natural selection, WHIM allele-inactivated cells were enriched over wildtype allele-inactivated cells. First, one copy of Cxcr4 in HSPCs was inactivated in vitro by CRISPR/Cas9 editing with a single guide RNA (sgRNA) that does not discriminate between WHIM and wildtype Cxcr4 alleles. We have now developed a two-step preclinical protocol of autologous hematopoietic stem and progenitor cell (HSPC) transplantation towards this goal. We previously reported that Cxcr4-haploinsufficient ( Cxcr4 +/o) hematopoietic stem cells (HSCs) have a strong selective advantage for durable hematopoietic reconstitution over wildtype ( Cxcr4 +/+) and WHIM ( Cxcr4 +/w) HSCs, and that a WHIM patient was spontaneously cured by chromothriptic deletion of the disease allele in an HSC, suggesting that WHIM allele inactivation through gene editing may be a safe genetic cure strategy for the disease. WHIM syndrome is an autosomal dominant immunodeficiency disorder caused by gain-of-function mutations in chemokine receptor CXCR4 that promote severe panleukopenia due to bone marrow (BM) retention of mature leukocytes. ![]() ![]() ![]() After the fireworks, she shows him her darkroom, portfolio and opens up about her mother flaking out, forcing her into the foster care system. One thing I loved about Samurai Deeper: Kyo was its invocation of actual Japanese history, and it galvanized me to study the Sengoku Jidai a bit in college. Kyo is a separate person, but his arch-rival sealed him within his own body. We find out relatively earlier that Kyo is not a split personality (I’m lookin’ at you, Stephen Grant/Marc Spector/Jake Lockely, who I will eventually be writing about), but is rather imprisoned within Kyoshiro due to some complicated past events. Where Kenshin had a secondary personality, however, the story with SDK is far, well, deeper. Kenshin has a suppressed secondary personality which is bloodthirsty and dangerous Kyoshiro and Kyo’s relationship starts off the same way - Kyo emerges to defend Kyoshiro and the bounty hunter who arrested him, Yuya Shiina, from dangerous threats. There is, to be sure, a similarity to Rurouni Kenshin in that way. The only catch is that Kyo shares a body with his arch-nemesis, the scruffy, mildly-criminal medicine peddler Kyoshiro Mibu. He’s a near-legendary samurai, famous for killing over one thousand enemies at the Battle of Sekigahara. He’s called “Demon Eyes” Kyo, on account of his bright, blood-red eyes making him look, well, like a demon. Set in the immediate aftermath of the Sengoku Jidai, SDK is about the titular character, Kyo. Let’s start with the basics - and, as always with my “Some Thoughts On” reviews, some spoilers will be inevitable: The manga was great, if imperfect, but the Anime was shockingly…Different. SDK became one of my favorite works of literature in my college years, but when its anime adaptation reached my eyes, well…I was disappointed. I branched off from Toonami classics like Rurouni Kenshin and Outlaw Star and landed on more esoteric things, such as Great Teacher Onizuka and - perhaps unavoidably - Samurai Deeper: Kyo, by Akimine Kamijyo.
